Duphaston - Duphaston is an orally active progestogen which acts directly on the uterus, producing a complete secretory endometrium in an estrogen-primed uterus. At therapeutic levels, Duphaston has no contraceptive effect as it does not inhibit or interfere with ovulation or the corpus luteum. Furthermore, Duphaston is non-androgenic, non-estrogenic, non-corticoid, non-anabolic and is not excreted as pregnanediol. Duphaston helps to regulate the healthy growth and normal shedding of the uterus lining. Therefore, it may be useful in the treatment of menstrual disorders such as absent, irregular or painful menstrual periods, infertility, premenstrual syndrome and endometriosis.

Indication: Used to treat irregular duration of cycles and irregular occurrence and duration of periods caused by progesterone deficiency. Also used to prevent natural abortion in patients who have a history of habitual abortions.

The active ingredient of Duphaston is Duphaston that is synthetic sex hormone with progestagens affect. Duphaston is similar to Progesterone. Progesterone is essential hormone for the development of endometrium(healthy womb lining) that is necessary for pregnancy. Duphaston contributes to beginning of secretory phase in endometrium, decreasing the risk of endometrium's hyperplasia or blastomatosis caused by estrogens. Duphaston is a progestogen hormone. The brand name is Duphaston and manufactured by Solvay Pharmaceuticals.

Duphaston was first introduced to the market in 1961, and is currently approved in over 100 countries world-wide. It has an estimated cumulative exposure of more than 28 million patients.

Duphaston is a potent, orally active progestogen indicated in a wide variety of gynaecological conditions. Although similar in molecular structure and pharmacological effects to endogenous progesterone. It is orally active at far lower doses. Its freedom from oestrogenic, androgenic, anabolic, corticoid and other undesirable hormonal effects gives it additional benefits over most other synthetic progestogens. The therapeutic use of Duphaston is closely related to its physiological action on the neuro-endocrine control of ovarian function, as well as on the endometrium. As such, it is indicated in all cases of relative or absolute endogeneous progesterone deficiency.

Duphaston has also been registered as hormone replacement therapy (HRT) to counteract the negative effects of unopposed oestrogen on the endometrium. Duphaston is safe and well-tolerated. The incidence of side-effects is remarkably low.

Hormone Replacement Therapy: To counteract the effects of unopposed estrogen on the endometrium (inner lining of the uterus) in hormone replacement therapy for women with disorders due to naturally or surgically induced menopause with an intact uterus. Progesterone Deficiencies: Treatment of progesterone deficiencies eg, dysmenorrhea (painful menstruation); endometriosis (growth of uterine tissues outside the uterus with associated symptoms); secondary amenorrhea (cessation of menstruation); irregular cycles; dysfunctional uterine bleeding ; premenstrual syndrome; threatened and habitual abortion associated with proven progesterone deficiency; infertility due to luteal (ovarian yellow body) insufficiency.

In cases where there is or is suspected a deficiency in endogenous progesterone or where the specific action of Duphaston has been clinically demonstrated eg, in infertility, threatened abortion, habitual abortion, dysmenorrhoea, endometriosis and irregular cycles. Moreover, combined with an oestrogenic substance, Duphaston can be applied in secondary amenorrhoea and dysfunctional bleedings.

Infertility due to luteal insufficiency; threatened abortion related to luteal deficiency; habitual abortion associated with proven progesterone deficiency; amenorrhoea (secondary); function uterine bleeding due to hormonal imbalance in the absence of organic pathology eg, submucous fibroids or uterine cancer; endometriosis; dysmenorrhoea (primary) unrelated to an obvious physical cause eg, uterine polyps, pelvic inflammatory disease or uterine leiomyomata. Adjunct to oestrogen replacement therapy.
